Abstract
This pilot project will determine if eicosanoids stimulate or inhibit the expression of cyclo- oxygenase-2 (COX-2) in mammalian macrophages. Murine macrophages (RAW264.7) will be stimulated to elicit COX-2 by challenge with lipopolysaccharide together with inflammatory cytokines in the presence or absence of exogenous eicosanoids including lipoxin A4, leukotrienes B4 and C4 or prostaglandin E2. The expression of COX-2 in these cells will be determined by (1) Western blotting of PAGE-separated lysates using monoclonal antibodies to COX-2 and (2) the appearance of COX-2 mRNA expression. Further experiments will also selectively inhibit the synthesis of different eicosanoids by RAW264.7 cells to determine their effect on COX-2 expression.
