Abstract
(1) Make new mutants in the myosin-binding region of Drosophila flight muscle actin by site- directed mutagenesis, then (2) put these, and some mutants already made, into flies by P- element transfection. (3) Study functioning of transformed flies' muscle by flight ability, of the muscles by fibre mechanics and gel separation of flight muscle proteins, and of the isolated mutant actin by affinity binding, in vitro motility/force assays, biochemical kinetic assays and NMR studies. (4) Analyse extant mutants also. (5) MLC-2 light-chain expression in E. coli will be done early in the grant. (6) Interpret the results in terms of the mechanisms of the cross-bridge cycle.
