Abstract
(a) To separate the contribution from (i) distorted cross-bridges (ii) detached cross-bridges to the phosphate released from ATP in phosphate- water oxygen exchange experiments in intact, skinned, muscle fibres. (b) To measure the effect of distortion on the rate constant of the force- producing step in the contractile cycle. (c) To determine the rate constant for the reverse of the hydrolysis step in the ATPase cycle by quenching the reactions occurring on the detached cross-bridges.
