Abstract

Our original discovery that many cells contain plasma membrane receptors for serum RBP has now been extended by the demonstration of specific membrane-bound receptors for urinary and nasal mucosa OBP. We aim to (i) clone the receptors for RBP using an expression cloning approach and use hybridisation methods to clone the presumably related OBP receptor, (ii) investigate using protein engineering and the construction of chimaeric proteins, the sites on RBP and OBP responsible for specific interaction with their respective receptors, (iii) study the cellular uptake process for retinol using expressed cellular RBP and (iv), use NMR to study the role and conformational changes of the active site Trp in the apo-holo RBP transition.
