Abstract

The aim of this work is to study the role of metabolites in the regulation of transcription of Calvin cycle genes in wheat and Arabidopsis. Northern slot blot analysis and PCR techniques will be used to determine the changes in expression of mRNA encoding FBPase, SBPase and PRKase in intact plants and protoplasts. Metabolite- responsive cis-acting elements will be identified using deletion mutants of the FBPase gene upsteam regions from wheat and Arabidopsis. This analysis will use a combination of transient expression in protoplasts and stably transformed transgenic plants. A combination of in vivo footprinting and in vitro gel-shift analysis will be used to identify and localise DNA-protein interactions.
