Abstract

We aim to map 100 T-DNA integrations carrying Ds elements in order to improve the efficiency of targeted insertional mutagenesis in Arabidopsis. New sAc and Ds elements would be constructed that allow transposition events to be selected in the greenhouse. Landsberg erecta transformants with single copy T-DNA integrations carrying the Ds element would be generated and plant DNA flanking the insertion isolated. The flanking DNA would be hybridized to arrayed YAC clones that form the physical map covering all five chromosomes. Targeted tagging experiments would be undertaken to provide information on the number of families that need to be screened to obtain a particular mutation.
