Abstract

The potential of simple sequence repeat (SSR) based markers will be evaluated for applications in fundamental and applied aspects of cereal genome research. SSRs will be isolated in parallel from barley (SCRI) and wheat (JIC) and characterised. Pairs of oligonucleotide primers will be designed to the unique flanking sequences and used to amplify the corresponding locus from total DNA. The polymorphic information content of each marker (PIC-value) will be calculated using a set of genotypes chosen to best represent the genetic diversity present in the respective primary gene-pools. Transportability will be examined using accessions from the secondary and tertiary gene-pools and from related cereals and grasses. Polymorphic SSRs will be mapped intrachromasomally to existing segregating populations. The generated information will be assembled into a standard format database to facilitate general access.
