Abstract
Recently we reported on a novel family of calcium-dependant calmodulin-binding transcription activators (CAMTAs; JBC 277: 21851; 2002). In the proposed research we plan to: (1) characterise CAMTA-responsive DNA elements using recombinant CAMTAs and nuclear extracts in vitro, and analysing chromatin/CAMTA complexes in vivo; (2) identify CAMTA-target genes using microarrays with RNA from camta knockout mutants; (3) employ bioinformatics tools to identify putative CAMTA-responsive elements; (4) study CAMTA-mediated gene expression in vivo using reporter genes, and the role of calcium in CAMTA regulation using aequorin (luminometry and imaging). We expect to elucidate a novel mechanism of calcium-mediated regulation of gene expression in plants.
