Abstract

This project will apply a new viral-based functional genomics approach, developed as SCRI, for the location of novel proteins associated with the plant Golgi apparatus, an organelle central to the processing of food materials in crop species. cDNA libraries from Arabidopsis tissues will be fused to the florescent protein and expressed en masse, using the viral-based vectors, in plants transformed with green fluorescent protein-tagged Golgi marker proteins. Lesions containing double-labelled Golgi will be excised, the cDNA sequenced and expressed via Agrobacterium infiltration in tobacco leaves. Proteins with confirmed association with the Golgi will be further characterised, to identify unique plant Gogli-specific and plant Gogli matrix proteins. Funds are also sought to establish a bioinformatics platform for dissemination of cDNA-GFP sequences.
