Abstract

Our DNA-array analysis of Fe/Fur (Fe uptake regulator) dependent gene expression in E. coli has identified 14 potential novel Fe-transporter genes, and a major new category of Fe-controlled genes encoding Fe-proteins involved in respiration which suggests a new iron-homeostatic mechanism. We now aim to perform a more extensive analysis of Fe/Fur regulation in E. coli and to identify the functions (or rationale for Fe regulation) of some of the more interesting Fe/Fur regulated genes identified by us so far: several predicted Fe transporters including one related to Ftr1p of yeast; a Cu exporter (the Cus system); a ribonucleotide reductase (NrdFEIH); and the Ni transporter. We also intend to characterise the Fe/Fur-controlled extracellular reductase activity of E. coli and investigate the role of the Suf system in scavenging cellular Fe during Fe restricted growth.
