Abstract

The aim is to identify the specific cell-types where genes responsible for high decarboxylase activity around veins are expressed and to determine the function of the gene products. We have recently found that in the vascular system of tobacco petioles activities of NAD-ME, NADP-ME and PEPCK are similar to those in the bundle sheath of C4 plants (Nature, 415: 451-454). Arabidopsis also has high activities of these enzymes around the vascular system and RT-PCR shows that only certain members of each gene family are expressed in these cells around the vasculature (Hibberd unpub). We will determine the exact cells in which each gene is expressed, investigate the mechanisms controlling expression of these genes and use isotope feeding and T-DNA insertional mutants to understand the function of the gene products.
