Abstract

Plant heterotrimeric G proteins are involved in responses to light, pathogens and hormones, yet the molecular context in which they function is not understood. In this proposal, the yeast two-hybrid Ras recruitment system will be used to identify proteins that interact with GPA1. Interactions will be tested between GPA1, a chorismate mutase isoform, and a putative regulator of G protein signalling protein. Interactions will be confirmed in vitro and characterised in biochemical assays and by ectopic over-expression. Mutants will be isolated in proteins that interact with GPA1 and double mutants with other signalling mutants such as gpa1 generated. The phenotype and response of these to known G protein mediated events will be determined.
