Abstract

Saprophytic cord-forming basidiomycetes play a central role in nutrient cycling in terrestrial ecosystems. We will define the complex patterns of N-redistribution in Phanerochaete velutina and Serpula lacrymans developing in a patchy resource environment using photon-counting scintillation imaging. The spatial pattern of expression of key genes involved in N- metabolism, particularly on the urea cycle, and N-regulation (areA) will be mapped using RT-PCR. Metabolite levels will be quantified using NMR in parallel. The role of the dynamic vacuolar network in long-distance N-transport will be determined using 4-D confocal microscopy and fluorescence recovery after photobleaching. Results will be integrated using an in silico simulation approach.
