Abstract

The E. coli FNR protein is a global regulator, controlling the expression of more than 50 genes, in response to oxygen availability. Promoters of target genes possess a sequence that closely matches the FNR binding site consensus (TTGATN4ATCAA). The E. coli genome sequence and advances in genomic technologies offer opportunities to gain new insight into the full extent of the FNR modulon and the cellular processes that are influenced by oxygen availability. It is proposed to analyse the genome sequence to identify all the possible functional FNR sites. Transcriptomics will then be used to test these predictions and thereby obtain a global view of the regulatory activities of FNR. New members of the FNR modulon with unusual promoter architectures will then be selected for detailed characterisation of their regulation and function.
