Abstract

Biofilm development will be quantified in Rhodobacter sphaeroides WS8 under a range of different environmental conditions using GFP as a marker and reporter. Confocal and fluorescence microscopy will be used to quantify development. The roles of motility, the chemotaxis (che) system and the quorum sensing (cer) system in biofilm development, maintenance and dispersal in R. sphaeroides will be determined using mutational studies. Genes involved in biofilm development will be detected using in vivo promoter trapping technology (RIVET) and these will be characterised. Six of these genes will be studied further in vivo, using mutational studies and GFP tagging, to elucidate their role in biofilm formation and development
