Abstract

F-box proteins recruit substrate proteins for ubiquitin (Ub)-mediated proteolysis. COI1 is an F-box protein required for jasmonate (JA)- regulated defence and fertility, and we predicted that it recruits regulators of JA responses to Ub-mediated proteolysis. Here we propose to identify proteins that are ubiquitinated by the COI1 Ub- ligase. Proteins that immunoprecipitate with epitope-tagged COI1, and proteins that immunoprecipitate with epitope-tagged Ub in wild plants but not coi1 mutants, will be purified and identified by mas spectrometry. From these candidates, substrates for COI1-dependent proteolysis will be identified by their more rapid disappearance, as GUS fusions, from wild-type plants than from coi1 mutants.
