Abstract

Our search for RNAi in Blumeria graminis hordei reveals evidence by inference, notably homologues of the Neurospora crassa quelling genes, qde 2 and qde 3. Here, we seek evidence by experiment by exploiting methanol inducible expression and subsequent silencing of GFP (which we have demonstrated in Pichia) and thence the silencing of the Bgh catB, cPKA and cutA genes. Each proposed gene to be silenced meets the qualifying criteria, notably, is single copy, shows evidence for involvement in differentiation and/or pathogenicity, carries a known RT-PCR transcript profile (deemed important for activation of the switch at specific time-points), and its protein product is detectable.
