Abstract

Transfection of pollen tubes with aequorin has detected a transient elevation of cytosol calcium during generative cell binary. Tobacco CDPK-7 expression is limited to late anthesis fission and pollen tube growth and fusion constructs indicate the protein kinase is specifically targeted to the male generative cell membrane. Using state of the art proteomics, the protein substrates and their amino acid phosphorylation sites for CDPK-7 in the generative cell membrane will be identified and phosphorylation status estimated during cytokinesis. Mis expression of CDPK-7 and photolysis of loaded caged calcium will be used to investigate the regulation of pollen tube cytokinesis by the cytosol calcium transduction pathway.
