Abstract

We propose to identify and characterise knockouts for 10 maize genes, that we have previously shown to have senescence-enhanced expression (Sees), by hybridisation to microarrayed Mutator-AFLP products. The 10 Sees have similarity to sequences within the database, however, in all cases their role in the senescence process is either unknown or unclear. Once plants with Mu-insertions within Sees have been identified they will be backcrossed before selfing and characterisation at the molecular, biochemical and physiological levels. The application of high throughput mutagenesis and detailed characterisation to cereal genes differentially expressed during leaf senescence will allow a greater understanding of the genetic role of this important developmental pathway and agronomic trait.
