Abstract

The nature of the interaction between the Brassica stigmatic S(incompatibility)-receptor kinase (SRK) and the male SI determinant (SCR) will be established by utilising in vitro binding assays and recombinant protein/peptide-array epitope mapping. SCR is one member of a family of pollen coat proteins (PCP-A class), previously identified by the applicant, which interact specifically with stigmatic S-locus linked or related proteins. In particular this programme will i) ascertain whether SRK and SCR interact directly ii) determine whether the interaction is modulated/stabilised by other pollen coat proteins such as PCP-A1 (which also binds SRK) and/or stigmatic S-proteins, and iii) identify key protein interaction and S-specificity domains of S-receptor complex members.
