Abstract
The RNA replicase complex of Cereal Yellow Dwarf Virus (CYDV) will be isolated and purified, and the virus- encoded and host-encoded protein components will be identified. Cis-acting sequences, essential for CYDV RNA replication, will be identified by mutational analysis using both in vivo and in vitro approaches. The role of a genome-linked viral protein (VPg) in priming RNA synthesis will be investigated. The intracellular membrane site of CYDV replication, and the protein(s) involved in replicase localization, will be determined by in situ RNA hybridization, immuno-electron microscopy and fluorescent microscopy.
