Abstract

Using genetic mapping and transient assay procedures the Ry PVY resistance gene has been localised to a 25kb region in the potato genome that does not contain typical NBS/LRR resistance genes. We shall investigate each of the four genes in the 25kb Ry region to find out whether they act either individually or in combination to mediate resistance. We shall characterise the gene(s) with the Ry function and investigate the nature of the interaction with the viral elicitor of resistance using yeast two hybrid and immuno- precipitation based methods. To assess the similarity of Ry and other resistance mechanisms we shall investigate and compare the components of the signalling pathways using chemical inhibitors and virus- induced silencing of candidate genes. Finally we shall investigate the functionality of Ry as a transgene in different potato cultivars and other solanaceae.
