Abstract

The sesquiterpene lactone (SL) profile in lettuce will be modified by sense and antisense expression of sesquiterpene cyclases. Altered profiles will be examined in transgenic cell suspensions and regenerated plants. The role of cyclases in the biosynthesis of guaianolide and germacranolide SLs including the phytoalexins lettucenin A and costunolide will be examined. The products of sesquiterpene cyclases cloned from lettuce will be determined and catalytic activity investigated using site-directed mutagenesis.
