Abstract

DNA ligases play crucial roles in DNA replication, excision repair and genetic recombination but our knowledge of these enzymes in plant systems is rudimentary. Recently we have cloned three distinct Arabidopsis DNA ligases including a novel DNA ligase unique to higher plants. We will employ a combined biochemical and molecular genetic approach to (i) dissect post transcriptional control mechanisms which determine DNA ligase I isoform expression in Arabidopsis, (ii) investigate the targeting of DNA ligases to intracellular sites of action and (iii) elucidate the role(s) of each DNA ligase in DNA metabolism.
