Abstract
We hypothesise that the S-derivatisation of protein thiols with the tripeptide glutathione (gamma-glu-cys-gly) is a profound and rapid mediator of cellular responses to redox changes in plants, directly modulating the activity of proteins involved in metabolism and cell signalling. Using Arabidopsis thaliana as a model system, we propose to identify the proteins which become S- glutathionylated following oxidative insult and characterise the effect of this modification on their activity. In preliminary studies we have demonstrated that a protein tyrosine phosphatase (PTP) activity is regulated by S- glutathionylation and emphasis will be placed on examining the link between redox change, S-glutathionylation and protein-phosphotyrosine metabolism.
