Abstract

Bacterial ATP binding cassette (ABC) transporters and their eukaryotic family members are involved in a variety of important house keeping and disease related functions. Although ABC transporters have been studied for over 30 years, the structure of these multidomain systems is unknown and the mechanism of transport is not understood. The widely studied ABC maltose transporter MalEFGK is used as a model to apply a combination of biochemical and genetic techniques to determine basic structural and functional properties and to address the important question of mechanism.
