Abstract

The structure of the 3' and 5' untranslated regions of the RNA of a fungal mitochondrial RNA virus (mitovirus) will be determined with chemical and enzymatic probes. The roles of 5'- and 3'- terminal structural elements in mitovirus RNA replication will be investigated by in vitro mutagenesis using both in vitro RNA synthesis with a template-dependent RNA polymerase and in vivo RNA synthesis. Template competition, and gel mobility shift experiments will be used to investigate interactions of the mitovirus RNA polymerase with RNA structural elements. Essential cis-acting sequences will be used to construct replicating mitochondrial vectors which could have widespread application in mitochondrial biology.
