Abstract

The research will enhance understanding of mechanisms for importing nucleus- encoded proteins into chloroplasts, and will focus on the outer envelope membrane Toc complex. The ppil mutant (lacking Toc33) is a unique resource (Science, 1998, 282: 100-103) that will be exploited fully. Four different approaches will be employed: (1) The import defect of ppil will be characterised in detail, elucidating the specific role played by Toc33; (2) Antibodies against Toc33 and Toc34 will be generated and used in immunoprecipitation experiments to investigate the composition of Toc complexes; (3) Reverse screens for mutants lacking other Toc proteins (Toc 34, 75, 86) will be conducted. Characterisation of single and double mutants will illuminate Toc functionality; and (4) A ppil screen will be conducted; this should identify new translocon components.
