Abstract

The COI1 gene is required for jasmonate (JA)-regulated defence and fertility in Arabidopsis. We isolated COI1, and predicted it codes for an F-box protein which recruits repressors of JA responses to ubiquitin-mediated proteolysis. To test this we used COI1 as bait in a yeast 2- hybrid screen of an Arabidopsis cDNA library, and isolated six interacting clones. Three are candidate repressors for COI1-targeted ubiquitination, and three are SKP1s, components of the ubiquitination complex. We propose to test the function of these COI1- interacting proteins in Arabidopsis, by down-regulation and by co-immunoprecipitation. We shall also identify other genes in the JA pathway by mutation, positional cloning, and by a new 2- hybrid screen.
