Abstract

Symptom expression in virus-infected plants involves a programmed pattern of changes in gene expression. We have previously identified many genes whose expression is regulated at a whole plant level during CaMV infection of Arabidopsis. Using micro- capillaries, we will sample RNA from individual cells, and measure abundance of individual viral and host mRNAs using RT-PCR. We will develop a novel macro-array hybridisation and measure simultaneously, the levels of multiple transcripts in single cells. Patterns of gene expression in symptomatic tissue will be determined. Virus variants, and host symptom- response mutants, will be used to determine the influence of virus and host genotype on virus-inducible alterations in gene expression.
