Abstract

Fimbriation is a substantial drain on the amino acid pool. Preliminary studies show that the phase variation of type 1 fimbriation in E. coli is stimulated by alanine and leucine, and that this regulation involves a change in the interaction of a regulatory protein (Lrp) with fim DNA. We plan to determine (i) how the amino acids affect Lrp to stimulate the site-specific recombination that causes phase variation (ii) why alternative Lrp-fim nucleoprotein structures affect switching so differently and (iii) the affect of nitrogen stress on fimbriation. This regulation appears to be a key control point for amino acid consumption and this study should illustrate how an important cellular structure is controlled in response to nutritional stress.
