Abstract
We have recently cloned a guard cell phosphoinositide-specific phospholipase C (PI- PLC) gene. When the expression of this gene is manipulated the phenotype of the resulting plants is consistent with an impairment in the ability of the guard cells to respond to extracellular stimuli. We wish to capitalise on these preliminary results and employ whole plant, cell physiological and molecular techniques to investigate thoroughly the contribution that PI-PLC makes to guard cell stimulus-response coupling. By characterising the response of these plants to stimuli known to influence stomatal turgor and gene expression (ABA, CO2, light, extracellular calcium) we shall determine which stimulus-response coupling pathways utilise PI-PLC. We shall also use ratio photometry to investigate the contribution that PI-PLC makes to defining stimulus- induced increases in cytosolic free calcium and hence address the question of specificity. In addition as there is evidence that the expression of plant PI-PLCs can be regulated by environmental signals we shall determine whether the expression of a PI-PLC gene is controlled by extracellular stimuli in guard cells.
