Abstract

A putative CA2+ channel has recently been sequenced as part of the Arabidopsis genome project. This gives us - for the first time - the opportunity to address the molecular basis of calcium signal generation in plants. The channel will be characterised functionally after heterologous expression and, in transient expression studies, assessed for its ability to evoke changes in cytosolic free CA2+ in response to stress stimuli. The expression pattern will be mapped to yield clues to function, and physiological function assessed more directly by overexpression and the analysis of knock-out mutants identified by reverse genetic approaches. The molecular basis of the regulation of channel activity will be assessed with mutants in which key regions of the protein thought to be involved in voltage- sensing and control by phosphorylation have been modified.
