Abstract

The objective of the present proposal is to be use the heterologous S. pombe alternative oxidase expression system to further investigate the structure of the alternative oxidase by site- directed mutagenesis experiments. The goal of these studies will be: 1) to investigate the active-site by mutating the residues postulated to act as ligands, 2) to investigate the function of the highly conserved surface-located helix, 3) to probe the nature of the quinone binding site and 4) to further characterise the quinone binding site using new alternative oxidase inhibitors.
