Abstract

Calcium signalling plays a key role in many aspects of fungal development, including tip growth in filamentous fungi. However, the pathways by which Ca2+ crosses the plasma membrane remain to be identified at a molecular level. We have recently demonstrated that the CCH1/MID1 genes of yeast are jointly involved in signalling-related Ca2+ entry, and have now identified homologs of these genes in Aspergillus nidulans. We will (i) obtain full- length clones of these genes; (ii) construct deletion and overexpression strains; (iii) characterise the phenotypes of these strains, including Ca2+ influx and cytosolic free Ca2+ homeostasis (using apoaeaquorin transformants); (iv) initiate structure-function studies on Aspergillus CCH1 with a view to understanding mechanisms of ion permeation and regulation.
