Abstract

We have identified a gene, rpoI, in Rhizobium leguminosarum the symbiont of peas which is involved in siderophore production and N2 fixation. The sequence of RpoI indicates that it is a member of the family of ECF sigma factors, being most closely related to the PvdS protein of Pseudomonas. Experiments are described to: (i) see if RpoI is functionally equivalent to PodS and if it is regulated by fur; (ii) use reporter fusion mutagenesis to see what genes, particularly those involved in symbiotic N2 fixation, are regulated by RpoI; (iii) demonstrate biochemically and genetically if RpoI is a sigma factor; (iv) characterise nodule developments in rpoI mutants; (v) study the factors that regulate rpoI expression.
