Abstract

Induction of fission yeast cdc25 transcripts in tobacco led to more, smaller lateral roots which comprised smaller cells compared with the wild type. The smaller cell size stabilised indicating that the Cdc25 phosphatase was down-regulated by plant cell cycle proteins. What are these regulatory proteins and how do they, and the Cdc25 phosphatase cause these developmental changes? Using cdc25 as bait in yeast two hybrid (2H) assays in Arabidopsis we shall isolate cDNAs encoding putative regulators of Cdc25. These will be used to transform Arabidopsis in which we will examine lateral root size and frequency. We shall also transform IAA (alf) mutants defective in lateral root morphogenesis to test the hypotheses that cdc25 is at the end of a signal transduction chain which initiates laterals. This would link cell cycle regulatory genes with an IAA-mediated developmental event.
