Abstract
Recent work has identified that the respiratory chain of the filamentous fungal pathogen, Ggt, includes both conventional and alternative oxidases as well as a novel b-type cytochrome. The initial aim is to identify the site of electron partitioning between the cytochrome and alternative pathways and assign functional roles to the alternative oxidase and the novel b-type cytochrome. Such work will employ the use of specific inhibitors to dissect the respiratory chain of isolated mitochondria to enable spectrophotometric, oxygen consumption and fluorescence imaging analysis to be employed. Sequence analysis and reconstitution assays will provide in-depth information as to the physico-chemical properties of the alternative oxidase, identify possible quinol binding sites and enable quantification of proton translocation.
