Abstract

The aims of this project are to determine the amino acid residues in the stigmatic S- protein of Papaver rhoeas that play a key role in the recognition of incompatible pollen using site-directed mutagenesis. A series of mutants will be constructed with the express objectives of: [A] Identification of S-protein mutants defective in receptor recognition. This will encompass: 1) defining the residues which interact with the recently identified accessory receptor (SBP)-binding region and 2) the residues that participate in S-allele specific pollen recognition. We also intend to complement the mutation analysis by undertaking: [B] structural analysis of a stigmatic S-protein, using crystallographic or NMR approaches, and [C] studies of the interaction of stigmatic S-proteins with the pollen S-receptor.
