Abstract

This proposal will initiate the characterisation of a unique family of haemolytic proteins by investigation of the properties and export of the E. coli hlyE gene product. The hlyE gene encodes a novel haemolysin and is a member of the FNR-modulon. Preliminary work has shown that active FNR is involved in both HlyE expression and export. The aims of this proposal are to: 1) determine the biochemical and biophysical properties of purified HlyE; 2) identify and study the regulation of the genes required for HlyE export under anaerobic conditions; 3) investigate structure- function relationships in HlyE in order to identify those regions of the protein associated with haemolytic activity and export.
