Abstract

Molecular isolation of the Mlo gene, a negative regulator of pathogen defence and cell death in barley, revealed related genes in monocot and dicot plant species. Mlo appears to be the founder of a novel and abundant family of plant regulatory proteins which exhibit structural similarities to mammalian G protein coupled receptors. We propose a thorough structural and functional analysis of Mlo by applying protein topology and subcellular localisation studies. A novel transient transformation system will be employed to identify functional domains in Mlo by using site-directed mutagenesis and domain swaps between family members. A series of pharmacological compounds will be used to test the involvement of heterotrimeric G proteins in mlo-mediated defence with a simplified coleoptile monolayer tissue system.
