Abstract

We shall exploit our breakthrough finding that dormant, non-growing cells of M. luteus can be resuscitated in growth medium only by the addition of picomolar concentrations of an autocrine proteinaceous growth factor (Rpf), which shows strong similarity to predicted gene products in Mycobacterium tuberculosis and Mycobacterium leprae. We shall do this by over-expressing this and a related M. luteus protein in E. coli and characterising them biochemically. We shall develop quantitative assays for their activity and establish their specificity and production kinetics, whether the secreted gene product is subjected to specific proteolytic cleavage, and whether any such cleavage leads to the modification of biological activity.
