Abstract

We propose to study the structure of higher plant photosystem I (PSI) using electron microscopy of two-dimensional (2-D) crystals of the complex. These crystals are the first reported for the higher plant complex, and they open up the possibility of obtaining much more structural data, allowing it's comparison with the simpler cyanobacterial PSI complex. A 3-D structure will be obtained for negatively- stained crystals, and then cryo-electron microscopy of unstained specimens will be carried out in order to extend the resolution of the structural data beyond 2.0 nm. Mapping of the locations of the higher plant PSI subunits will be attempted by antibody labelling; salt and chaotrope washing; and by the use of various mutants. The binding sites for soluble electron transfer proteins on the complex will also be investigated by electron microscopy.
