Abstract

Transgenic Arabidopsis expressing cauliflower mosaic virus protein P6 from 3 different virus strains exhibit a range of symptom-like phenotypes, and provide a model system for investigating host-virus interactions in symptom expression. Seeds from transgenic lines will be mutagenised, and the progeny screened for mutants with altered phenotype severity. We aim to identify mutants that do not map in the transgene, i.e. in host loci controlling or involved in symptom expression. We have already identified one such mutant, which is insensitive to P6. This, together with further mutants, will be characterised genetically, and where possible cloned and sequenced. Differential display PCR will be used to elucidate the effects of gene VI expression and virus replication on patterns of host gene expression. The effects on the patterns of gene expression within the mutants will be analysed.
