Abstract

The potato virus X mediated expression system will be used to target green fluorescent protein (GFP) to the endomembrane system of tobacco leaf cells. Mobility of GFP tagged with KDEL, various calreticulin constructs and other vacuolar storage proteins will be assessed by recovery from photobleaching of brefeldin A induced sheets of endoplasmic reticulum. This will produce novel information on the mode of retention/retrieval of ER luminal proteins. This work will be coupled with the localisation of the GFP-tagged K/HDEL receptor to investigate recycling of KDEL proteins to and from the cis-Golgi. The ability of GFP-tagged mammalian Golgi transferases to partition into plant Golgi will be investigated and transgenic lines produced expressing fluorescent Golgi. These will be used in studies of Golgi dynamics in living plant cells.
