Abstract

As part of our goal in understanding the dynamics of the plant actin cytoskeleton we have identified a novel 35kDa phosphoprotein that copurifies with the maize actin depolymerising factor, ZmADF3; either with the ZmADF3 alone or with the ZmADF3:actin complex. We will use an immunoaffinity column procedure to isolate ZmADF3 complexes from maize suspension culture cell extracts. The 35kDa protein and corresponding cDNA will be isolated. In addition, we will use the sensitive two-hybrid system for identifying clones from a maize suspension culture cDNA library encoding proteins that specifically interact with ZmADF3. All identified proteins will be tested for functional activity in ZmADF3/G-actin binding assays, ZmADF3/F-actin disruption assays and, where appropriate, phosphorylation assays.
