Abstract
The Papaver pollen/SI system is becoming a classic model system for investigating second messenger signalling in intact plant cells. We recently described the first Ca2+ waves in plant cells. They may be initiated by increases in [Ca2+]i and [Ins(1,4,5)P3]i, and propagated by InsP3-induced Ca2+ -release (IICR) and modulate pollen tube growth. We have established that the self-incompatibility (SI) response is mediated by [Ca2+]i and recently obtained evidence for Ca2+ wave formation. We propose to establish the spatial and temporal dynamics of these Ca2+ waves in intact living pollen using Ca2+ -imaging. We aim to establish whether this involves extracellular and/or intracellular Ca2+ and to identify Ca2+ stores and Ca2+ channels involved. We will investigate the signalling pathways involved in [Ca2+]i modulation in the SI response and establish if there is a role for IICR and/or Ca2+ - induced Ca2+ -release (CICR).
