Abstract
We aim to determine the physiological function of the widespread periplasmic cytochrome c', which we hypothesise to buffer NO in vivo. Insertional inactivation of the genes for cytochrome c' from Paracoccus denitrificans and Rhodobacter capsulatus will be followed by a detailed physiological analysis of the mutant phenotypes, with emphasis on the involvement of the cytochrome in NO metabolism. In addition to physiological analysis in batch and continuous liquid cultures, the toxicity of NO and O2 will be assessed on plates by disk diffusion susceptibility assays. Whole cell EPR spectroscopy will be used to examine cytochrome c' coordination state in vivo. Kinetic experiments are outlined which will examine whether cytochrome c' is a NO buffer and/or whether it reacts with O2. Also, genetic loci which confer resistance to NO will be isolated from random genomic libraries.
