Abstract

A novel disease resistance signalling gene, EDS1, has been cloned in the model plant, Arabidopsis. It is required for resistance to an oomycete pathogen, Peronospora parasitica, mediated by several different RPP genes, and for another gene, RPS4, conferring resistance to a bacterial pathogen, Pseudomonas syringae. EDS1 also contributes to resistance to P. parasitica isolates that normally only infect Brassica plants. The project proposes a functional analysis of EDS1 with respect to its sequence, expression pattern, activity, and molecular interaction with already cloned resistance gene products, including RPP5, which is known to be EDS1-dependent. An interaction trap cloning strategy will be employed to isolate other proteins that bind to EDS1 and will be complemented by a genetic screen for additional mutations in the EDSI-signalling pathway.
