Abstract

This project has two complementary aims. The first is to identify genes with important functions in the formation of infection structures in a plant pathogenic fungus. The corresponding genes will be isolated by cloning the genetically tagged mutational insertions and their function assessed by a combination of molecular genetic and ultrastructural analysis. The second aim is to develop a novel technique to generate gene disruptions in plant pathogenic fungi. The techniques will use a transposon engineered to carry a fungal selectable marker. This will be used to mutate plasmid-borne genomic sequences which can then be introduced directly into fungal strains to carry out directed gene disruptions. This will allow genes identified based on their expression, or the location of their protein products, to be rapidly tested to determine their role in pathogenicity.
